I've told this before, but the nearest big town to where I grew up had a Sears catalog store with the "SEARS" letters individually lit. Sometimes it would say "EARS," which is comedy gold when you're five. So my inner five-year-old thinks this is terrific.
Then there was the Shell station that used to be along I-26 that, back in the day, had the five letters individually lit on the big sign. One night my dad, who used to drive trucks at night, saw it when the "S" was burned out....

The Steak House restaurant in Myrtle Beach one had a problem with their sign that made them "The Steak Ho."

What Tom describes was the butt of a Jay Leno headline where it was the Outback Steakho.
There was another where what was supposed to be a Big Lots resulted in BIG OTS.

I once saw an ANGUS BURGER sign with the G missing.
Didn't want me none of that.

@Homer: Exactly!
Another: When I lived in Florida, there was a Discount Auto Parts nearby that had a sign malfunction, so for a few days it was Disco Auto Parts.

Years ago there used to be a PICK-A-FLICK video store in one of the strip malls on Decker Blvd. Thanks to the character set they used on the sign, on first glance, the word 'flick' took on a whole different meaning!!!!

I was in Camden not too long ago when there was a Shoney's that certain letters were illuminated and others weren't. Here's the kicker, the letters that weren't illuminated on one side were on the other.
